چکیده

Recognition of conventional foreign antigen by T cells is determined by the expression of multiple variable regions of both ol and 3 chains of the T cell receptor (TCR) oe/3 heterodimer. In contrast, there exists a class of antigens that appears to interact with the TCR c~/3 heterodimer through the variable region on the 3 chain (V3), independent of other TCR components, a property that has led to their designation as superantigens. The goal of the present study was to analyze Voe use in V36 + T cells responsive to the superantigen, Mla ~. Results indicate that while deletion of T cells expressing V36 in Mlsa-expressing mice is essentially complete and therefore appears to occur regardless of Vol usage, in vitro Mls ' stimulation of T cells from Mlsa-negative mice results in significant skewing of Voe use among responding V36 + T cells. This indicates that Vot expression influences recognition of the superantigen, Mls" by mature peripheral T cells.

Analysis of the interaction site for the self superantigen Mls-1a on T cell receptor V beta

Superantigen bound to major histocompatibility complex (MHC) products have been shown to stimulate T cells in a V beta-specific manner. Mouse T cells bearing V beta 8.1 usually respond to the self superantigen, Mls-1a, whereas T cells bearing V beta 8.2a do not. Previously, using site-directed mutational analysis, we identified the residues of natural variants of T cell receptor (TCR) V beta 8....

Unique molecular surface features of in vivo tolerized T cells.

Differential expression of surface markers can frequently be used to distinguish functional subsets of T cells, yet a surface phenotype unique to T cells induced into an anergic state has not been described. Here, we report that CD4 T cells rendered anergic in vivo by superantigen can be identified by loss of the 6C10 T cell marker. Inoculation of Vbeta8.1 T cell antigen receptor (TCR) transgen...

Influence of T cell receptor V alpha expression on Mlsa superantigen- specific T cell responses

Recognition of conventional foreign antigen by T cells is determined by the expression of multiple variable regions of both alpha and beta chains of the T cell receptor (TCR) alpha/beta heterodimer. In contrast, there exists a class of antigens that appears to interact with the TCR alpha/beta heterodimer through the variable region on the beta chain (V beta), independent of other TCR components...

Expression of bacterial superantigen genes in mice induces localized mononuclear cell inflammatory responses.

Bacterial superantigens are potent T cell activators, and superantigen proteins have been injected into mice and other animals to study T cell responses in vivo. When superantigen proteins are injected, however, the T cell stimulatory effects cannot be confined to specific tissues. Therefore, to target superantigen expression to specific tissues, we used gene transfer techniques to express bact...

Major histocompatibility complex-specific recognition of Mls-1 is mediated by multiple elements of the T cell receptor

We have recently shown that recognition of the mouse mammary tumor virus 9-associated superantigen (vSAG-9) by murine V beta 17+ T cells is strongly influenced by the major histocompatibility complex (MHC) class II haplotype of the presenting cells, resulting in a form of MHC-restricted recognition. This finding was unexpected, because T cell recognition of another well-characterized retroviral...
